Depending on the website hosting your article, there are various exit points your readers can leave your online article page. There are links to other collection of articles, link leading to the website menu and many others. You can't help it if they choose to go those ways. But what you can do if they do click on your affiliate link.
Most writers put their affiliate product sales page directly linked to the article. This is fine but you are relying 100% that they buy the product for you to benefit. If they don't, you won't have any clue on who they are and how you can contact them.

After keying in their email (with name if required) they are then taken to your affiliate website. At this point, even if they decide not to buy your product, you already have the information to contact them through your auto responder.
Your guess is right, if you keep on promoting your affiliate product in your auto responder and also the link back to your article or website if you have one. You can also include other related products, if you have.
As they sign up voluntarily and there is an option of not receiving the email, you can be rest assured that they are your target audience as long as they are in the list.
